cI under control of the plac promoter. BBa_R0010 and BBa_P0151 The composite part is made of pLacI promoter (which is inducible with IPTG), a weak RBS sequence, a cI gene that codes for cI repressor from E. coli phage lambda and two terminators. This operon produces a regulatory protein that represses cI sensitive promoters.

Usage and Biology

cI is a transcription inhibitor of bacteriophage Lambda. Also known as Lambda Repressor, cI is responsible for maintaining the lysogenic life cycle of phage Lambda. It has been adapted to regulate gene expression and is used in molecular biology. It is also modified with an LVA tail for rapid degradation of the protein. This biobrick was used to characterize BBa_K195613 – a GFP expressing device that is under control of cI sensitive promoter in vivo .Obtained data indicates that this biobrick (BBa_K077039) might be prone to high unpredictability of protein output, due to combined effects of low translation levels of cI protein (weak RBS) and the instability (LVA tail) of the protein.
